The longer the primer strongest interaction with the target DNA, since more hydrogen bonding occurs between the complementary bases. Increased specificity for the same reason. The longer the primer further information is stored in its sequence, more than able to recognize the correct sequence corresponding to the target DNA. Increase interaction associated with multiple primers led to greater stability at high temperatures, so that the higher annealing temperature.
